ABSTRACT

Objective:To establish a candidate reference method for the determination of angiotensin Ⅱ in human plasma by isotope dilution liquid chromatography tandem mass spectrometry (ID-LC-MS/MS) and to evaluate its performance.Methods:Using [ 13C 6- 15N]-angiotensin Ⅱ as the internal standard, the plasma was accurately weighed by gravimetric method and mixed with a certain amount of internal standard. At the same time, enzyme inhibitor was added. After zinc sulfate solution protein precipitation and reversed-phase solid-phase extraction plate treatment, it was analyzed by liquid chromatography tandem mass spectrometry. The multi reaction ion monitoring mode(MRM)was selected by mass spectrometry to detect specific ion fragments of angiotensin Ⅱ and internal standard. The linearity, sensitivity, precision, recovery rate and uncertainty of the performance of the established method were evaluated according to ISO15193. Results:Angiotensin Ⅱ had good linearity in the range of 10-1 000 pg/g ( r=0.999 5), the lower limit of quantification was 7.68 pg/g, the analytical recoveries were 97.14% to 102.85%, intra-batch imprecision≤3.21%, inter-batch imprecision≤2.96%, and total imprecision≤3.67%. Conclusion:A method for the determination of plasma angiotensin Ⅱ was established by ID-LC-MS/MS. The method is accurate and reliable, and is expected to be a reference method for the determination of plasma angiotensin Ⅱ.

ABSTRACT

Objective:To evaluate the reliability and validity of the Chinese version of the Orebro musculoskeletal pain questionnaire (OMPQ-CHN) using patients with non-specific lower back pain.Methods:The OMPQ was translated into simplified Chinese according to the Beaton cross-cultural translation guidelines, and then 186 patients with acute or subacute lower back pain were asked to respond to its questions. Based on the results, the internal consistency and test-retest reliability were quantified, and its validity was assessed using content, criterion-related and predictive validity.Results:The Cronbach′s α for the instrument was 0.804. Its ICC was 0.833 (95% CI: 0.683-0.915). I-CVI ranged from 0.86 to 1, S-CVI/Ave=0.99. The correlation between the OMPQ-CHN and other questionnaires ranged from r=0.419 to r=0.646. The AUCs predicting pain, dysfunction and sick leave ranged from 0.723 to 0.810. Conclusions:The OMPQ-CHN has good reliability and validity, and suitably assesses the psychosocial risk factors of Chinese patients with low back pain.

@#Objective To explore the intra- and inter-rater reliability of shear wave elastography (SWE) for the patellar tendon and quadriceps femoris stiffness.Methods From October to November, 2017, the stiffness of the patellar tendon and quadriceps femoris of 20 healthy men was measured by SWE, and measured again five days later. The results were evaluated with interclass correlation coefficient (ICC).Results For patellar tendon, ICC=0.79 in intra-rater and inter-rater; for rectus femoris, ICC=0.71 in intra-rater and 0.73 in inter-rater; for vastus lateralis muscle, ICC=0.84 in intra-rater and 0.74 in inter-rater; for vastus medialis,ICC=0.95 in intra-rater and 0.94 in inter-rater.Conclusion It is a reliable and repeatable method to measure the stiffness of patellar tendon and quadriceps femoris by SWE.

ABSTRACT

Objective To investigate the effect of measuring value transfer for human serum samples assigned by the reference laboratory network on improving the trueness of seven enzyme activities in clinical laboratories,such as ALT,AST,GGT,LDH,CK,AMY and ALP.Methods Depending on the medical imtitutions at all levels contacted by 5 reference laboratories in North China,South China,East China and Southwest China,the corresponding clinical laboratory measuring value transfer/traceability network was established.The frozen human serum samples with good interehangeability and standard material characteristics,including calibrator,sample 1 and sample 2,were provided by Beijing Aerospace General Hospital,and were assigned by 5 reference labotatories in four regiom.These samples were sent to 48 clinical laboratories.These clinical laboratories measured sample 1 and sample 2 according to their standard operating procedures,and then measured.the two samples again after adjusting their measurement system by using the supplied calibrator.The changes of trueness of detection results in these laboratories were evaluated according to the WS/T 403-2012 standard,and the changes of consistency for ALT and AST before and after measuring value tramfer were investigated.Results The results of AMY,ALP,GGT,CK and LDH calibrator,sample 1 and sample 2 assigned by the established network were 138.7 U/L,278.5 U/L and 68.3 U/L,265.3 U/L,94.5 U/L and 134.4 U/L,195.8 U/L,89.0 U/L and 158.9 U/L,393.7 U/L,260.0 U/L and 645.3 U/L,and 302.0 U/L,250.0 U/L and 452.7 U/L,respectively.The percentages of sample 1 and sample 2 which met the bias requirements of the WS/T 403-2012 standard before measuring value transfer for AMY,ALP and GGT were 65.9% and 61.0%,76.6% and 78.7%,and 66.7% and 70.8%,respectively,while after measuring value transfer,they were 89.2% and 83.8%,86.7% and 80.0%,and 85.4% and 91.7%,respectively.The percentages of sample 2 which met the bias requirements of the WS/T 403-2012 standard before measuring value transfer for CK and LDH were 64.6% and 58.3%,respectively,while after measuring value trander,they were 93.5% and 84.8%,respectively.The coefficients of variation (consistency) of sample 1 and sample 2 for ALT and AST before measuring value tramfer were 12.9% and 11.3%,and 10.2% and 8.9%,respectively,while after measuring value transfer,they were 9.3% and 8.2%,and 5.6% and 5.9%,respectively.Conclusion The calibration of routine measurement systems based on the measuring value transfer for human serum samples assigned by the reference laboratory network may improve the comparability of 7 enzyme actvities measurement results in chnical laboratories at all levels obviously,which deserves to be further spread.

Objective To assess the reliability of strength of shoulder rotator cuff using a hand-held dynamometer among subjects with spinal cord injury. Methods 20 spinal cord injury subjects without shoulder pain participated in this study. A hand-held dynamometer was used to assess the strength of shoulder rotator cuff. Two therapists (A/B) obtained the strength of shoulder rotator cuff. One of the therapists assessed again 5days later. The intra-and inter-reliability (ICC) of strength of shoulder rotator was calculated. Results The strength of rota-tor cuff was assessed by the hand-held dynamometer. The intra-reliability of external and internal muscle strength was 0.94 and 0.95, respec-tively. The inter-reliability was 0.91 and 0.93, respectively. Conclusion A hand-held dynamometer is a feasible and reliable tool to assess the strength of rotator cuff among subjects with spinal cord injury.

ABSTRACT

Objective To investigate the reliability of the morphologic measure of the iliotibial band (ITB) with musculoskeletal ultra-sound imaging. Methods 20 healthy young subjects were measured with the musculoskeletal ultrasound imaging of the thickness of bilateral ITB at the levels of the femoral condyle by 2 testers, and one of the testers measured again 3-5 days later. The intraclass correlation coeffi-cient (ICC) and minimum detectable change (MDC) were calculated, and the thickness of ITB in both sides was compared. Results The ICC of test-retest was 0.89 (R) and 0.85 (L), and it was 0.82 (R) and 0.84 (L) of inter-testers. The MDC was 0.41-0.51 mm. There was no signifi-cant different between right and left sides for the ITB thickness among healthy subjects (P=0.97). Conclusion Musculoskeletal ultrasound imaging is a feasible and reliable to measure the ITB thickness among young healthy subjects.

@#Objective To assess the perception of rehabilitation therapeutics students to their learning environment.Methods The Dundee Ready Education Environment Measure (DREEM) inventory was used to measure rehabilitation therapeutics education environment by investigating two hundred and forty-seven baccalaureate rehabilitation therapeutics students.Results The rehabilitation therapeutics students perceived the total education environment and four subscales which included perceptions of learning and teachers, environment, and students' academic self-perceptions as relatively satisfactory. Moreover, they perceived that the teachers were knowledgeable, the relationships between teachers and students were better, and this school was well regulations. But the scores of students' social self-perceptions was lower (15.23 ±4.16). Especially, there were some problems in learning approach and accommodations.Conclusion To take advantage of competent teachers, good teacher-student relationship, and proper regulations and overcome unfavorable factors in social activities and approaches to learning is a guarantee of good rehabilitation therapeutics education environment.

ABSTRACT

@#Objective To observe the effect of hot spring hydrotherapy and routine rehabilitation on spasticity of patients with incomplete thoracic spinal cord injury (SCI). Methods 25 patients with incomplete thoracic SCI were divided into hydrotherapy group (n=15) and control group (n=10). The hydrotherapy group received both routine rehabilitation and hydrotherapy,and the control group only underwent routine rehabilitation.They were evaluated with Modified Ashworth Scale (MAS) before and after treatment. Results There was a significant reduction on MAS in both the groups after treatment (P<0.01), and it was more in the hydrotherapy group (P<0.05). Conclusion The hot spring hydrotherapy can facilitate the release of spasticity after incomplete thoracic SCI.
